Genetic Variation and Pathogenicity of a Pseudorabies Virus Variant Strain in Mice

Abstract: This study aimed to understand the molecular genetic variation and virulence characteristics of porcine pseudorabies virus (PRV) in China. The main glycoprotein gene sequences (gB, gC and gE) of PRV ZJ strain isolated from a pig farm inoculated with Bartha-K61 vaccine were determined. and its pathogenicity to 6-week-old mice was studied. Phylogenetic analysis results showed that this strain was highly homologous to epidemic strains in China after 2012, and had the genetic characteristics of the mutant strain. However, there was a unique change of S510G in the amino acid position 510 of gE gene, which was different from the mutant strain. ZJ strain was highly pathogenic to BALB/c mice, and the challenged mice showed typical neurological symptoms. The mortality rate reached 100% on day 5 after the mice were infected with 100 LD50. This study laid a foundation for further exploration of the variation characteristics and pathogenesis of this virus.
