Comparative Analysis of Transcriptome Levels in Ovarian Tissues of Yunnan Dulong Chicken and White Leghorn Chicken

Abstract: The aim of this study was to screen and analyze differentially expressed genes in ovarian tissues of Yunnan Dulong chicken and White Leghorn chicken using RNA-seq technology, and to lay the foundation for subsequent studies on gene function, breed improvement, and germplasm characteristics of Dulong chicken. In this study, the ovarian tissues of three 43-week-old Dulong chickens in good health were collected for transcriptome sequencing and compared with the ovarian tissue of White Leghorn Chicken in similar rearing conditions and age. The results showed that there were 1 273 differentially expressed genes in Dulong chickens compared with White Leghorn chicken, among which 522 genes were up-regulated and 751 genes were down-regulated. GO and KEGG enrichment analysis showed that differentially expressed genes were mainly involved in proteolysis, translation, exosomes, membrane components, binding, and catalysis, and played an important role in protein synthesis, ECM-receptor interaction, oxidative phosphorylation, adhesion plaque, and other pathways. Significant enrichment pathway analysis showed that Dulong chickens had certain advantages in egg quality and immune resistance, but there was still a certain gap between Dulong chickens and White Leghorn chickens in laying performance. RPL22, ATP5I, COX5A, JAKMIP1, CATH2 screened in this study were mainly enriched in the pathways related to resistance, egg-laying performance, and egg quality, which could be used as important candidate genes for further study.
