Expression of 10 ku Protein Gene from Taenia solium and Its Preliminary Application

Abstract: Recombinant expression plasmids, designated as pGEX-4T-TSO10 and pGEX-4T-CE10, were constructed by insertion of the TSO10 gene from Taenia solium oncosphere and the CE10 gene from the larva, respectively, and identified by sequencing. The results of SDS-PAGE, Western-blot and lamellar scanning showed that both fusion target proteins expressed in E. coli were 35 ku in molecular weight and can be recognized by the positive serum from a cysticercosis patient, and that their contents accounted for 25% and 30% of total bacteria proteins, respectively. Furthermore, the results of ELISA indicated that the recombinant proteins are suitable for diagnosis of cysticercosis.
