氧化应激对断奶仔猪色氨酸分解代谢的影响

Effects of Oxidative Stress on Tryptophan Catabolism in Weaned Pigs

摘要: 旨在研究氧化应激对断奶仔猪色氨酸分解代谢产物及关键酶活性的影响。采用单因子试验设计,选用24头28±1日龄断奶长白×大白仔猪,分别饲养在24个猪栏中,经过预饲后随机分为3个处理组,每个处理组8个重复,每个重复1头猪。氧化应激组:仔猪腹腔注射10 mg·kg-1体质量Diquat,自由采食;正常对照组:仔猪腹腔注射无菌生理盐水,自由采食;采食配对组:仔猪腹腔注射无菌生理盐水,控制采食量与氧化应激组仔猪相同。所有仔猪自由饮水,试验期7 d。结果表明,与对照组和采食配对组相比,氧化应激组:(1)氧化应激降低了仔猪生长性能,极显著降低血清SOD(超氧化物歧化酶)(P<0.01)和GPx(谷胱甘肽过氧化物)活性(P<0.01),血清MDA(丙二醛)含量极显著增加(P<0.01);(2)氧化应激降低了血清Trp(色氨酸)水平,显著或极显著增加了血清Kyn(犬尿氨酸)含量(P<0.05或P<0.01)及Kyn/Trp比值(P<0.01),血清5-HT(5-羟色胺)含量显著增加(P<0.05);(3)氧化应激显著增加了肝脏TDO(色氨酸2,3-加双氧酶)总活性(P<0.05),全酶(P<0.05)和酶蛋白活性(P<0.05),对十二指肠IDO(吲哚胺2,3加双氧酶)活性无显著影响。结果提示,Diquat诱导的氧化应激主要通过增强肝脏TDO活性来增加断奶仔猪色氨酸分解代谢。

Abstract: This study was designed to evaluate the effects of Diquat-induced oxidative stress on tryptophan catabolism in weaned pigs. According to a single factorial arrangement, a total of 24, Landrance ×Yorkshire weaned piglets were assigned into three treatments of 8 animals each, consisting of pigs given ad libitum access to feed (control), challenged with diquat (10 mg·kg-1 BW) and given ad libitum access to feed (oxidative stress), or pair-fed to receive the same amount of feed as the Diquat-challenged pigs (pair-fed). The trial lasted for 7 days. The results showed that: ⑴ The growth performance and activities of SOD (superoxide dismutase) (P<0.01) and GPx (glutathione peroxide dismutase) (P<0.01)significantly declined in Diquatchallenged pigs, which followed by increased serum malondialdehyde (MDA) (P<0.01); ⑵ Diquat-induced oxidative stress decreased serum free Trp (tryptophan), augmented serum Kyn (kynurenine) (P<0.05 or P<0.01), Kyn/Trp (P<0.01) and 5-HT (5-hydroxytryptamine) (P<0.05); ⑶ After Diquat injection, the liver TDO (tryptophan 2,3-dioxygenase) activities including total activity(P<0.05),holoenzyme(P<0.05) and apoenzyme activity(P<0.05) were enhanced, the effect of oxidative stress on IDO (indolamine 2,3dioxygenase) was no significant difference. These results suggested that Diquat-induced oxidative stress increased catabolism of tryptophan by augmenting TDO activity in weaned pigs.
