Sequence Characteristics and Expression Analysis of CA5B Gene in Pigs

Abstract: Abstract: (Objective)This study aimed to analyze the expression characteristics of CA5B gene of Tongcheng and Landrace pigs, and to analyze the effects of CA5B on the tissue development and metabolism of pigs.(Method) We Used pig transcriptome data, and analyzed the tissue expression profile of CA5B gene and the spatiotemporal expression of skeletal muscle at 27 development time points. Then we conducted the CA5B protein conservative analysis, phylogenetic tree, and protein interaction network analysis by the CA5B protein sequence database in NCBI. Finally, Targetscan, PicTar, miRanda and other software were used to analyze the miRNA bound to CA5B, and the editing site of CA5B was analyzed.(Result)The results showed that CA5B is highly expressed in pig fat, testis and ovary, and the expression in embryonic phase during the growth and development of skeletal muscle is higher than that in postnatal period. CA5B gene was highly conservativeexpressed in mice, humans and pigs. Phylogenetic tree found CA5B in mouse is earlier than that in humans and pigs. Interaction network showed CA5B expressed protein interacts with mitochondrial transmembrane transport protein, myosin-like coiled-coil protein, cytochrome family and other proteins. In addition, CA5B may be Regulated by ssc-miR-22-5p through sequence alignment (p<0.05), and there are 3 potential RNA editing sites in CA5B.(Conclusion) The results suggest that CA5B may function in the mitochondrial respiratory chain and has an important role in the organization of energy storage and utilization.
