EGF、Gln和pGRF基因质粒对早期断奶仔猪肠道发育的影响

Effect of Epidermal Growth Factor, Glutamine and pGRF Gene Plasmid on the Intestinal Development of Early Weaning Piglets

摘要: 为研究表皮生长因子(EGF)及其与谷氨酰胺(Gln)和pGRF基因质粒的不同组合形式对早期断奶仔猪肠道发育影响的差异,选取同日龄断奶仔猪30头,等分到EGF、EGF+Gln、EGF+pGRF基因质粒、EGF+pGRF基因质粒+Gln和对照组(生理盐水)共5个处理中,每个处理3个重复,每个重复2头猪,进行对比试验,试验期21 d。试验结束时,从每个处理的每个重复中随机选取1头仔猪,空腹称重后屠宰取样。结果表明:EGF与pGRF基因质粒、Gln的不同组合形式对肝脏、胰脏和胃主要消化器官重量没有显著影响(P>0.05),但均不同程度促进了仔猪小肠的发育。其中,以EGF+Gln+pGRF基因质粒的效果最为明显,小肠、空肠前部、空肠中段、空肠后段及整个空肠的重量分别超过对照组37.12%(P<0.1)、103%(P<0.05)、86.27%(P<0.05)、100%(P<0.1)和96.02%(P<0.05)。空肠绒毛长度提高89.68%(P<0.1),细菌移位率为零。但在本试验中,各处理对黏膜刷状缘碱性磷酸酶的活性都没有明显影响(P>0.05)。

Abstract: An experiment was conducted to study the effect of epidermal growth factor(EGF),glutamine(Gln) and porcine growth release factor gene plasmid(pGRF gene plasmid)on the development of the digestive system in weaned piglets. In this trial, thirty piglets(Landrace × Large white × Taihu, initial weight 4.51 kg),weaned at 28 days, were randomly allocated to five treatment, three replicates of two piglets each. Pigs in different group were given different combination of the substances metioned above. Group I was given EGF alone; Group Ⅱ was given EGF and Gln; Group Ⅲ was given EGF and pGRF gene plasmid; Group Ⅳ was given EGF, Gln and pGRF gene plasmid; Group Ⅴ was given physiological saline as control. Samples were collected 21 days later. The weight of main digestive organs including stomach, liver, pancreas, small intestine and mucous of intestine were measured. The length of villi, the value of ALP in mucous of small intestine and the value of bacterial shift were detected. The results showed that the effect of the combined of EGF, Gln and pGRF gene plasmid was much better than that of EGF. Compared with control group, the weight of small intestine, frontal jejunum, middle jejunum, rear jejunum and entire jejunum of group Ⅳ were inereased by 37.12%(P<0.1),103%(P<0.05),8627%(P<0.05),100%(P<0.1) and 96.02%(P<0.05) respectively. In group Ⅳ, the length of villi in jejunum was increased by 89.68% and no bacterial translation was found. But the activity of alkaline phosphate enzyme wasn’t influnced significantly in every treated groups (P>0.05).
