Detection of Distribution of Porcine Circovirus Type 2 in Experimentally InfectedPiglets with Immunohistochemical Method

Abstract: 28-day-old healthy conventional piglets were inoculated nasally with porcine circovirus type 2 (PCV2) BF strain, the piglets were necropsy on different day post inoculation. Tissues were collected for preparing tissue sections and immunohistochemistry protocol were developed to detect distribution of PCV2 antigens in tissue samples. Results showed that in lymphoid tissues such as lymph node, spleen and tonsil were positive for PCV2 antigen. PCV2 positive signals also presented in heart, liver, lung, kidney, stomach, duodenum, cerebrum and cerebellum. Other tissues were negative for PCV2 antigen. Viral antigens were localized predominately in the cytoplasm of the infected cells and rarely observed in the nucleus of the cells. Results indicated that infection indeed presented in the tissues of the experimental piglets.
