The Development of PCR-RFLP for Detecting Deficiency of Uridine Monophosphate Synthase

Abstract: Deficiency of uridine monophosphate synthase (DUMPS) is a monogenic autosomal recessive disorder in cattle, resulting in early embryonic death of homozygous offspring. To identify the mutation responsible for DUMPS, a direct genome DNA test based on PCR-RLFP was developed. Altogether 224 dairy cows from 6 herds in Jinan and 53 Holstein bulls were analysed, of which 2 cows, i.e. 0.89% were confirmed DUMPS carriers in cows, and zreo in bulls. Experimention tested the method was repeatable, sensitive and stable.
