Polymorphism Analysis of MHC-DRB1 Gene in Chinese Merino Sheep by PCRRFLP

Abstract: The polymorphism of the exon 2 of MHC-DRB1 was investigated by nested PCR-RFLP technique at three loci digested with SacⅠ,Hin1Ⅰand HaeⅢ,respectively, in a total number of 211 individuals of Chinese Merino(Xinjiang Army Wasteland Reclamation) sheep, and genotypic frequencies and allelic frequencies were counted. The results showed that the extensive polymorphisms of the exon 2 of MHC-DRB1 were found at three loci digested with SacⅠ,Hin1Ⅰ and HaeⅢ,respectively, in Chinese Merino sheep, these sites were controlled by two, two and six alleles, respectively. As some base mutations were found in MHCDRB1 locus, and according to the results in this paper, twentyfour alleles were found in Chinese Merino(Xinjiang Army Wasteland Reclamation) sheep. The results of χ2 test showed that genotypes of MHC-DRB1 digested with SacⅠand Hin1Ⅰexcept HaeⅢ(P<0.01)fitted with Hardy-Weinberg equilibrium(P>0.05)
