Detection and Localization of the Viral RNA in Bovine Tissues Experimentally Infected with Foot-and-mouth Disease Virus with in situ Hybridization

Abstract: RNAs of Foot-and-mouth disease virus in bovine tissues from six experimentally infected cattle was localized and detected by in situ hybridization. Results showed that strong positive staining was viewed in four bovine tongue epithelial tissues 7-28 days post-infection. The strong positive staining particles were observed in the epithelial tissues of larynx and pharynx 7-21 days postinfection. The positive staining was not observed in the tongue epithelial tissues from 35 days post-infection cattle and the normal control cattle. No positive staining was viewed in the tissues from tonsils, lymph nodes and cuneus ungulae of all six experimental cattles. Results indicated that epithelium of tongue, larynx and pharynx were infected by foot-and-mouth disease virus. The results are very valuable and helpful to further elucidate persistent mechanisms of foot-and-mouth disease virus in cattle.
