Development and Evaluation of an ELISA Based on Synthetic Peptide for the Detection of Footandmouth Disease Virus Antibody

Abstract: The specific synthetic peptide of footandmouth disease virus(FMDV) main protective antigen VP1 was synthesized by a solidphase method, and was conjugated with carrier protein BSA. An ELISA for detecting FMDV antibody was developed and evaluated by using the conjugated peptide as the coating antigen. The sensitivity and specificity of it were 95.12% and 100%. Comparing this assay with two commercial ELISA kits by detecting 199 serum samples,results showed that the agreement of it with UBI FMD VP1 ELISA kit and liquid phase blocking ELISA kit was 98.49% and 96.98%, respectively. The results indicated that the ELISA using synthetic peptide as coating antigen is specific, sensitive, stable and easy, and can be used to monitor antibody level of FMD.
