POMC Expression and Localization Analysis in Different Coat Color Alpaca Skin

摘要：

旨在研究POMC基因对黑色素细胞的形成、发育以及色素生成方面的重要作用。本研究通过qRT-PCR方法比较不同毛色羊驼皮肤中POMC mRNA的表达水平，采用免疫组织化学技术对POMC蛋白的定位和表达进行蛋白水平分析。结果显示：POMC在棕色和白色羊驼皮肤组织中均有表达，且棕色羊驼皮肤中的POMC mRNA相对表达量为白色羊驼皮肤的23倍（P＜0.01），POMC蛋白表达部位主要在表皮层和毛囊的毛根鞘周围的角质形成细胞内，且POMC在两部分的表达显示棕色羊驼中的表达量高于白色羊驼（P＜0.05），表明POMC可能参与羊驼毛色的形成。

Abstract:

As a color-related gene，POMC plays an crucial role in the formation，development，pigmentation and other aspects of melanocytes.To study the expression level of POMC mRNA and localization of POMC protein in alpaca skin with different coat colors，quantitative real-time PCR (qRT-PCR) and immunohistochemistry were performed.The results showed that the quantity expression of POMC mRNA in brown alpaca was 23 times than in white alpaca (P＜0.01)，and POMC were expressed in root sheath and epidermis in both brown and white alpaca skin，moreover，the average protein expression of POMC in brown alpaca skin was higher than in white alpaca skin on the average optical density (P＜0.05).So POMC may be involved in the coat color formation.
