敖汉细毛羊毛囊FGF10、FGF18和FGFR3基因差异表达的研究

Study on FGF10,FGF18 and FGFR3 Differential Expression in Hair Follicle of Aohan Fine-wool Sheep

摘要:

本研究旨在解析FGF家族及其受体基因在细毛羊不同部位皮肤、不同时间mRNA和蛋白的表达及与细毛羊毛囊生长发育的关系,为揭示毛囊生长发育的分子机制奠定理论基础。首先通过基因芯片技术,筛选敖汉细毛羊毛囊发育的差异表达基因,并通过qRT-PCR对基因芯片结果的准确性进行验证。之后对得到的差异表达基因作进一步分析,可知FGFs家族在颈部和腹部,羊毛毛囊的生长旺盛期和缓慢期之间的mRNA表达量存在差异的有FGF10、FGF18和FGFR3基因。最后通过二维凝胶电泳(2-DE)和质谱技术(MS),对毛囊发育的差异表达蛋白进行筛选,得出FGFs家族蛋白质在颈部和腹部、羊毛毛囊的生长期和缓慢期之间仍存在差异表达的有FGF10、FGF18和FGFR3蛋白。结果显示,FGF10 mRNA在腹部表达量高于颈部,推测可能对毛囊的发育具有负调控作用;其蛋白表达量颈部均高于腹部,推测可能促进毛囊生长发育。FGF18在腹部其mRNA表达量旺盛期高于缓慢期,推测其可能维持此处毛囊的不活跃状态;其蛋白在颈部表达量均低于腹部,推测可能对毛囊的生长具有抑制作用。FGFR3 mRNA和蛋白表达水平无统一趋势,可能与毛囊的生长发育存在较复杂的关系。综合以上,FGF10、FGF18和FGFR3 mRNA和蛋白水平在毛囊生长发育的不同部位和不同时期存在差异表达,这提示它们可能与毛囊的生长发育具有一定的关系,并在不同水平发挥调控作用。研究结果为解析FGFs家族成员调控毛囊生长发育的分子机制奠定理论基础,为进一步加快细毛羊育种提供重要候选基因。

Abstract:

The objectives of the present study were to identify the FGFs mRNA and protein differentially expression at different stages of hair follicle growth and development,and also to clarify the relationship between them.Firstly,gene chips were used to screen the differentially expressed genes in hair follicle at different stages and parts,FGF10,FGF18 and FGFR3,belonging to FGFs family,were identified.The qRT-PCR were used to verify the accuracy of gene chips results.Then by two-dimensional gel electrophoresis (2-DE) and mass-spectrometric technique (MS) technologies,differentially expressed proteins were identified,including FGF10,FGF18 and FGFR3 proteins.The results showed FGF10 mRNA expression level was higher in abdomen than that in neck,suggested that it might play the negative regulation role in hair follicle development,and the expression of FGF10 protein was adverse.FGF18 mRNA expression level in abdomen was higher at anagen stage and protein expression was lower in neck,suggested it might inhibit the growth of hair follicles.FGFR3 had no consistent rules.These results showed that the expression of the 3 genes at mRNA and protein levels are different in hair follicle at different stages and parts,which indicate the 3 genes are related to the growth and development of hair follicle.The results may provide basic theory for understanding molecular mechanisms of FGFs regulating hair follicle growth and development,and also provide candidate genes for fine-wool breeding project.
