O型口蹄疫病毒O/LZ株拯救

摘要: 以构建的口蹄疫病毒O/LZ 株全长cDNA为模板,使用T7RNA 聚合酶系统在体外进行转录,得到病毒RNA。用脂质体转染法将转录物RNA导入BHK细胞,可观察到典型的口蹄疫病毒致细胞病变效应。对收获的病毒分别用RT-PCR、免疫荧光以及免疫电子显微镜观察等方法进行鉴定,结果表明拯救到特定的口蹄疫病毒。同时用空斑试验法观察了拯救病毒及其母本毒株的生长特性,结果表明二者没有明显差异,这将为进一步探索猪口蹄疫病毒致病的分子机制及研制新型口蹄疫疫苗奠定良好的基础。

Abstract: The cloned cDNA derived from the genome of FMDV O/LZ strain was used as template and was transcripted to produce RNA with T7 RiboMAXTM Express Large Scale RNA Production System. The RNA was transfected into BHK cells. The typical cytopathic effect which were caused by rescusing FMDV was observed. Furthermore,the rescusing FMDV were verified by RT-PCR,IFA and observation of immunoelectron microscope. The results suggested that the FMDV were successfully rescued. This study have paved the way for exploring vaccines against FMDV,pathogenic mechanism and so on.
