鹅Pit-1基因部分序列多态性分析

Analysis on the Polymorphism of Sequences of Pit-1 Gene in Goose

摘要: 根据鸡Pit-1 mRNA 和基因组序列设计1对引物,以籽鹅、狮头鹅、皖西白鹅、四季鹅、浙东白鹅和朗德鹅6个群体的DNA为模板,扩增鹅Pit-1基因的部分片段。结果发现该片段存在2个插入/缺失突变,共出现3种基因型,分别为AA、AB和BB;与AA基因型相比,BB基因型分别在扩增片段的132和145 bp后插入3和13 bp。所有群体在该突变位点上的等位基因频率均处于哈代温伯格平衡状态(P>0.05);朗德鹅群体基因型分布与其他5个鹅群体均存在极显著差异(P<0.01);籽鹅和狮头鹅群体内基因型的分布与其他3个鹅群体存在显著差异(P<0.05)。在狮头鹅和朗德鹅群体内,等位基因A为优势等位基因,而在皖西白鹅、四季鹅和浙东白鹅群体内等位基因B为优势等位基因。

Abstract: Genetic polymorphism of goose Pit-1 gene fragment was analyzed in 6 goose populations. The primer was designed according to chicken Pit-1 mRNA and genomic sequence. Two insertion/deletion mutations were found in the sequence of Pit-1 gene, and 3 genotypes AA, AB and BB were detected in 6 goose populations. Compared with AA genotype, there were 3 and 13 bp insertions in BB genotype after position 132 and 145 bp,respectively. All the populations were in Hardy-Weinberg equilibrium at this polymorphic site (P>0.05). The genotype distribution in Landosie was significantly different with that in other goose populations (P<0.01). The genotype distribution in Zi goose and Shitou goose were significantly different with that in other populations(P<0.05). Allele A was the dominant allele in Shitou and Landoise goose populations, while allele B was the dominant allele in Wanxi White goose, Siji goose and Zhedong White goose populations.
