全反式维甲酸对大鼠前体脂肪细胞增殖与分化的影响

Effects of All Trans Retinoic Acid on Proliferation and Differentiation of Rat Preadipocytes

摘要: 利用MTT比色检测不同浓度的全反式维甲酸(all trans Retinoic Acid ATRA)对大鼠前体脂肪细胞增殖的影响,采用油红O染色和吖啶橙染色观察大鼠前体脂肪细胞分化的形态变化;采用油红O染色提取法分析不同浓度ATRA对大鼠前体脂肪细胞分化的影响。MTT比色结果显示,低浓度(10-8~10-7 mol/L)的ATRA对前体脂肪细胞的增殖具有促进作用(P<0.01),而高浓度(10-6~10-4 mol/L)的ATRA则抑制前体脂肪细胞的增殖(P<0.01)。油红O染色和吖啶橙染色结果表明,前体脂肪细胞分化成脂肪细胞后,细胞内充满脂滴,由梭形变成椭圆形;油红O染色提取法结果显示高浓度(10-6~10-4 mol/L)的ATRA能够显著抑制前体脂肪细胞的分化(P<0.01),而低浓度(10-8~10-7 mol/L)的ATAR却显著促进前体脂肪细胞的分化(P<0.01)。

Abstract: ATRA has widely biological effects, which can regulate the proliferation, differentiation and apoptosis of cells. Therefore, the purpose of the experiment is to investigate the function of ATRA on the proliferation and differentiation of rat preadipocytes. In the present study, the effects of ATRA on proliferation and differentiation of preadipocytes was detected by MTT and quantitation of Oil Red O staining respectively; the morphologic changes of preadipocytes differentiation were analyzed by Oil Red O staining and Acridine Orange staining. The results of MTT show that the low concentrations (10-8~10-7 mol/L) of ATRA accelerate the proliferation of preadipocytes, but the high concentrations(10-6~10-4 mol/L)of ATRA inhibit it(P<0.01). The results of quantitation of Oil Red O staining show that the high concentrations(10-6~10-4 mol/L)of ATRA block the differentiation of preadipocytes, but the low concentrations(10-8~10-7 mol/L) of ATRA promote it(P<0.01). The staining pictures of Oil Red O and Acridine Orange demonstrate that when preadipocytes differentiate into adipocytes, they become oval and fill with lipid.
