PPAR基因多态性与肉鸡脂肪性状的相关性研究

Relationship between Genotype of PPARα and Body Fat Traits in AA Broiler Line

摘要: 以AA肉鸡为试验材料,采用PCR-SSCP技术,应用5对引物,对过氧化物酶体增殖剂受体(PPARα)基因的编码区进行了SNP检测,结果发现了1个多态性位点,使试验鸡群表现出EE、EF和FF 3种基因型。利用SAS软件(8.02版),统计分析了3种基因型与AA肉鸡的腹脂等屠体性状的相关性,结果表明:与EE基因型相比,FF基因型有较高的腹脂重和腹脂率,且差异显著(P<0.05),但FF基因型与EF基因型之间差异不显著;EF基因型比FF基因型有较高的腹脂重和腹脂率,但差异不显著(P>0.05)。试验结果揭示:PPAR基因对鸡的脂肪性状的代谢具有重要作用,可以作为分子标记用于鸡脂肪性状的分子标记辅助选择。

Abstract: Among the genes that are involved with the development of growth and development of fat tissues, PPAR subfamily members are one cluster of the most important genes. The object of this study were to identify polymorphism of PPARα gene in a AA broiler line, develop PCR-SSCP methods to detect those DNA polymorphism and evaluate the relationship between those polymorphism PPARα genes and body fat traits. The single nucleotide polymorphism (SNP) were found in exon regions of the PPARα. The differential genotype detected by PCR-SSCP were defined as EE, EF and FF. The statistics analysis results showed that the PPARα polymorphism was significant related to the abdominal fat(AF)weight and abdominal fat percentage(AFP)(P<0.05), and the chicken with the genotype of FF has a higher AF and AFP than that of EE; while there was no significant difference between the genotype of FF and EF. The chicken with the genotype of EF has a higher AF and AFP than that of EE, but did not attain the significant difference level (P>0.05). The result indicated that the PPARα play an important role in chicken fat tissue metabolism. The significantly correlation between PPARα and development of fat tissue suggested that this gene is a good marker for using in marker associated selection program.
