高原超细型细毛羊经济性状的微卫星标记研究

Study on Economic Traits of Alpine Ultrafine Wool Sheep Using the Microsatellite DNA Technique

摘要: 利用3对微卫星引物对70只中国美利奴高原型超细类群个体进行DNA多态性分析,结果表明:3个微卫星座位具有高度多态性,多态信息含量平均为0.739 1,有效等位基因数4.441 9。3个微卫星标记与经济性状间的相关分析表明,微卫星MCM218的160 bp等位基因和BMC1009的288 bp等位基因与中国美利奴高原型超细群体绵羊羊毛细度有极显著的相关性,相关系数分别为-0.956 1和-0.849 1。微卫星BMS1248的145 bp的等位基因与中国美利奴高原型超细型绵羊周岁体重有显著的相关性。相关系数为0.952 3。这一结论初步说明,微卫星MCM218的160 bp等位基因和BMC1009的288 bp的等位基因与中国美利奴高原型超细群体周岁绵羊羊毛细度存在一定程度的连锁,微卫星BMS1248的145 bp的等位基因与中国美利奴高山型超细型绵羊周岁体重也存在一定的连锁关系,为进一步分析细毛羊主要经济性状的连锁标记以及克隆相关基因打下基础。

Abstract: The study was designed to carry out DNA polymorphism analysis of a total of 70 Alpine utra-fine wool sheep (Chinese Merino) using 3 pairs of microsatellite primers. The results showed that 3 microsatellite Loci of the Alpine ultrafine wool sheep had high diversity, with the average polymorphism information content of 0.739 1 and the number of effective alleles of 4.441 9. The result of the correlation analysis between genetic diversity of microsatellite DNA and economic traits showed that both 160bp alleles of MCM 218 and 288bp alleles of BMC 1009 were highly correlated with wool fitness (P<0.01), with a correlation coefficient of -0.956 1 and -0.849 1 respectively; 145bp alleles of BMS1248 were also highly correlated with yearling weight(P<0.05), with a correlation coefficient of 0.952 3. This indicates that there is a linkage between the genetic diversity of microsatellite DNA and economic traits of Alpine ultra-fine wool sheep. Therefore, this is a fundamental study for further looking for markers of both linkage and cloning, and for early selection of the important economic traits of Alpine ultrafine wool Chinese merino.
