Abstract: In order to investigate the effect of castration on expression of TTF-1 and TTF-2 genes, realtime fluorescence quantitative PCR was applied to investigate the developmental expression patterns of TTF-1 and TTF-2 genes from the thyroid tissue of castrated and uncastrated Jinhua pigs on the age of 60, 90 and 120 days. The results showed that the expression of TTF-1 and TTF-2 genes increased slightly in uncastrated pigs, while they were relatively stable in castrated pigs. The expression level of TTF-1 and TTF-2 mRNA in castrated Jinhua pigs on 60 and 90 days were higher than that of uncastrated pigs, while the expression level of TTF-1 and TTF-2 on 120 days were lower than that of uncastrated pigs, but no significant difference (P>0.05). The results indicated that the real-time PCR method was faithful for detecting the expression analysis of TTF-1 and TTF-2 genes, and the expression of TTF-1 and TTF-2 mRNA increased after castration. Furthermore, there was a significant correlation between TTF-1 gene expression and meat quality traits.
