Construction of Eukaryotic Expression Plasmids Encoding Chicken IL-18 Gene and Study on Its Immunoenhancement on Newcastle Disease Vaccine

Abstract: Complete chicken IL-18 gene were amplified firstly by RT-PCR from spleen cells of chicken embryo inoculated with Newcastle disease virus. The gene were inserted into pcDNA3 vector, resulting in the construction of pcDNA3-IL18 plasmid. Immunoenhancement effects of the pcDNA3-IL18 plasmids on Newcastle disease vaccine were observed by the test of HI antibody, proliferation response of the T lymphocytes from thymus and the B lymphocytes from bursa of Fabricius. The results indicated that chicken IL-18 have obvious immunoenhancement effect on Newcastle disease vaccine. It can not only enhance the cell mediated immune response obviously,but also enhance the humoral response,thus improve the protective rate of ND vaccine.
