用单胚mRNA差显技术克隆山羊早期胚胎发育相关基因

摘要: 用单胚构建的mRNA差异显示技术,对体外培养的山羊早期2、4、8-16细胞期胚胎的基因表达进行了研究,并选择了一条在4细胞期胚胎特异表达的条带进行分析。结果表明:该片段与牛胰岛素样生长因子结合蛋白3(IGFBP3)基因具有88%的同源性。该基因通过调控IGFs的生物学活性而影响早期胚胎的生长发育,并具有促进细胞分裂和影响内分泌的作用,是羊早期胚胎发育过程中的重要调控因素。

Abstract: Single preimplantation embryo mRNA differential display was used to study the gene expression of 2-cell、4-cell and 8-16-cell stage goat embryos cultured in vitro. A specific fragment expressed in 4-cell stage was selected for further analysis. The result suggested that this fragment displayed high homology (88%) to cattle insulinlike growth factor binding protein-3 (IGFBP3) gene. IGFBP3 was the regulator of IGFs bioactivity, it can also promote cell division and influence endocrine activation. IGFBP3 was an important factor in the process of goat preimplantation embryo development.
