牛尿苷酸合酶缺乏症PCRRFLP检测方法的研究

摘要: 本研究通过PCR-RFLP建立了直接利用基因组DNA检测尿苷酸合酶缺乏症(DUMPS)突变的方法。对济南市6个奶牛场224头奶牛及53头荷斯坦种公牛进行了DUMPS的PCR-RFLP检测,共检出2头杂合母牛(携带者),杂合子占检测母牛群的0.89%,在荷斯坦公牛中只检测到一种基因型,没有发现隐性突变基因的携带者。实验证明,该方法重复性好、灵敏性高、稳定性可靠。

Abstract: Deficiency of uridine monophosphate synthase (DUMPS) is a monogenic autosomal recessive disorder in cattle, resulting in early embryonic death of homozygous offspring. To identify the mutation responsible for DUMPS, a direct genome DNA test based on PCR-RLFP was developed. Altogether 224 dairy cows from 6 herds in Jinan and 53 Holstein bulls were analysed, of which 2 cows, i.e. 0.89% were confirmed DUMPS carriers in cows, and zreo in bulls. Experimention tested the method was repeatable, sensitive and stable.
