小尾寒羊BMPR-ⅠB基因的多态性、效应及连锁分析

Polymorphism, Effect and Linkage Analysis of BMPR-ⅠB Gene in Small Tailed Han Sheep

摘要: 骨形态发生蛋白受体ⅠB(BMPR-ⅠB)的Q249R(A746G)突变使Booroola Merino绵羊的繁殖性能提高。该突变在小尾寒羊群体中也有分布。本研究用PCR-SSCP技术对299只小尾寒羊BMPR-ⅠB基因突变位点的多态性进行了检测,BB、B+和++的基因型频率分别为0.541 8,0.364 5和0.093 7,突变等位基因B的基因频率为0.724 1。对2个小尾寒羊羊场的111只有产羔数记录的小尾寒羊BMPR-ⅠB的Q249R突变对产羔数的效应进行了分析,估计出突变等位基因B对小尾寒羊产羔数的效应为:替代效应和显性效应,在第一胎分别为0.46和0.16,在第二胎分别为0.56和0.11; 在第一胎和第二胎BB比++分别多产0.77和1.02羔。对BB基因型小尾寒羊和微卫星标记OARJL36的连锁关系进行了分析,发现在BB基因型小尾寒羊, BMPR-ⅠB的B等位基因与OARJL36位点之间仍然存在重组。

Abstract: The Q249R mutation (A746G) in bone morphogenetic protein receptor-ⅠB (BMPR-ⅠB) is associated with the high prolificacy of Booroola Merino sheep. In our previous study, we found that, in Small Tailed Han sheep population, a high proportion of ewes carry such mutation. The present study further analyzed the polymorphism, effect and linkage of BMPR-ⅠB gene in this breed. The results indicated that the genotype frequencies of BB, B+ and ++ were 0.541 8, 0.364 5 and 0.093 7 respectively, and the allelic frequency of B was 0.724 1.The substitution effect and dominance effect of the B allele were estimated to be 0.46 and 0.16 for the first parity and 0.56 and 0.11 for the second parity, respectively. Moreover, the fully FecB-associated microsatellite marker OARJL36 was genotyped in the BB ewes of Small Tailed Han sheep. We found that, in Small Tailed Han sheep, recombination between BMPR-ⅠB gene and OARJL36 locus are still occurring.
