猪源和鼠源脑心肌炎病毒分离株基因组的比较分析

Comparative Analysis of the Genomes of Encephalomyocarditis Virus Isolates from Porcine and Mouse Origin

摘要: 为了分析猪源和鼠源脑心肌炎病毒(EMCV)基因组的差异,对分离自同一猪场猪临床样本和鼠组织样本的2株病毒(GX0601和GX0602)进行了全基因组序列测定和比较分析。结果显示,2个分离毒株的基因组全长(包含poly A)分别为7 729和7 725 nt,核苷酸和氨基酸同源性均在998%以上。与国外毒株及国内已报道的分离株的序列比较结果表明,2个毒株与BJC3和HB1的核苷酸同源性为9818%~9941%,推导的氨基酸序列同源性为995%~997%,与国外分离株的核苷酸序列和氨基酸序列的同源性分别介于8053%~9957%和931%~995%。基于基因组开放阅读框推导的氨基酸序列的系统进化分析表明,我国分离毒株与国外毒株PEC9、CBNU、BEL2887A/91和EMCVR的同源性较高,位于同一个亚群。研究结果提示,猪源与鼠源毒株的基因组具有很高的同源性,鼠是猪场EMCV感染的传染来源。

Abstract: The complete genomes of two strains of encephalomyocarditis virus (EMCV) (GX0601 and GX0602), isolated from clinical tissue samples of pig and mouse in a pig farm in Guangxi province, were sequenced and analyzed in order to investigate the differences between the genomes of viruses from different host origin. The results showed that the genome sizes of the two viruses were 7 729 and 7 725 nt, respectively, including poly (A) tail. The complete genomic nucleotide and amino acid identity between the two viruses were above 998%. Compared with foreign and domestic strains, the two viruses had 9818%9941% identity in nucleotide and 995%997% identity in deduced amino acid with BJC3 and HB1, while 8053%9957% identity in nucleotide and 931%995% identity in deduced amino acid with foreign strains. Phylogenetic tree based on the amino acid sequences of the entire ORF revealed that the Chinese isolates clustered together with PEC9,CBNU,BEL2887A/91 and EMCVR. Our results revealed that the complete genome between the viruses from pig and mouse origin had higher homology, suggesting that mouse might be the source of EMCV infection in pig farm.
