不同钙磷比例对体外培养番鸭破骨细胞生成及骨吸收功能的影响

Effects of Different Ratios of Calcium to Phosphorus on the Formation and Activation of Duck’s Osteoclasts Cultured in vitro

摘要: 从1~7日龄番鸭长骨骨髓分离破骨细胞(Osteoclasts, OC)进行培养,培养过程中添加不同摩尔浓度比例的钙磷双因子(CCa∶ CP为2∶1、1∶1、1∶2)。倒置显微镜观察细胞形态,于培养第1、3天进行抗酒石酸酸性磷酸酶染色(TRAP),培养7 d后扫描电镜观察象牙片吸收陷窝,研究不同钙磷比例对番鸭OC生成及骨吸收功能的影响。结果显示,培养第3天时试验组TRAP阳性细胞多于对照组,差异极显著(P<0.01)。试验组中除CCa∶ CP = 1∶1组与CCa∶ CP = 2∶1组之间无显著差异外(P>0.05),其余各组间差异均极显著(P<0.01)。扫描电镜观察,象牙片吸收陷窝程度与钙磷比例大小(CCa∶ CP为2∶1、1∶1、1∶2)成反比。证实合适比例的钙、磷(CCa∶ CP = 2∶1)通过抑制OC生成和活化,抑制OC的骨吸收活性。

Abstract: Osteoclasts (OC) were isolated from long bones of 1 to 7 days old ducks. In the culture process, different concentration ratios of calcium (Ca) to phosphorus (P) (CCa : CP = 2∶1,1∶1,1∶2) were added to the culture system. The formation and activation of these OC were detected by phasecontrast microscopy, histochemical staining for tartrate-resistant acid phosphatase (TRAP) staining at the 1st and the 3rd day, and detection of lacunar resorption through scanning electron microscopy at the 7thday. For three days culture, the number of OC in experimental group was more than control group very significant. There was no difference between CCa∶ CP = 1∶1 group and CCa∶ CP = 2∶1 group.The difference among other groups was very significant at the 3rd day. The results demonstrated that CCa∶ CP = 2∶1 inhibits bone absorbing activity by inhibitng the formation and activation of OC.
