肉种鸡饲养方式对子代胚肝蛋白表达谱的影响

Effect of Maternal Rearing on the Proteins Profiles of Liver in Offspring Broiler during Embryonic Development

摘要: 挑选40周龄体况良好的产蛋三黄肉种鸡200只,随机分为对照组(笼养)和试验组(平养),饲喂相同的肉种鸡日粮。收集受精种蛋各300枚并孵化,分别取9、14、19胚龄和出壳时的肝脏,经2DPAGE分离后,对经胶体考马斯亮蓝G250染色的图像用PDquest73软件进行差异表达分析,取差异表达的蛋白质斑点进行胶内酶切和MALDITOFMS分析。结果表明,试验组和对照组在9、14和19胚龄时,胚质量和肝质量均无显著差异;出壳时,试验组胚质量、肝质量均显著小于对照组(P<005)。蛋白表达谱分析发现29个在试验组和对照组间表达量存在两倍以上差异的蛋白斑点,其中22个得到鉴定。在这些被鉴定的蛋白中,大多涉及到机体内基础代谢的酶、能量产生及信号传导等。

Abstract: Fortyweek healthy Sanhuang (SH) broilers were randomly divided into floorreared (treatment) and cagereared groups (control), and feed with the same diet. During the experimental period, 300 fertile eggs were randomly collected from each group and directly incubated in an electric forceddraft incubator. Livers were collected on day9, 14, 19 of incubation and the hatching, respectively. Proteins were extracted and separated by 2DPAGE and stained with colloidal Coomassie Brilliant Blue. Then, the stained gels were scanned and analyzed using PDQuest73 software. The differential protein spots with ingel trypsin digestion were subsequently identified by MALDITOFMS. The results indicated that the embryo and absolute liver weight of floorreared group was significantly lower than that of the control at hatching, although no significant differences were observed in both embryo and absolute liver weight between the floorreared group and the control on day 9, 14 and 19 of embryonic development. The result of offspring broiler liver protein profiles showed that twentynine proteins had expressed at least 2.0fold variance compared to the control during embryonic development and twentytwo of them were identified. Most of these proteins were highly related to carbohydrate metabolism, energy production and signal transduction.
