Sequence Variations on the Bovine Insulinlike Growth Factor-I Gene

The polymorphism of the insulin-like growth factor 1 (IGF-I) gene in Nanyang, Luxi cattle, and Chinese Simmental was analyzed by PCRSSCP and PCRRFLP. The results showed that the polymorphisms were not detected by PCRSSCP and PCR-RFLP in the 5′flanking region, and the exon 5 of the IGF-I gene. And three genotypes, AA, AB, and BB, were detected in the exon 2 of the IGF-I gene. The polymorphism fragments were cloned and sequenced. The sequencing results indicated that there were three single nucleotide mutations: A→G at 35 bp, C→T at 248 bp, A→G at 256 bp in the exon2 of the IGF-I gene, but the mutations at 35 bp and 248 bp were missense mutations. The mutation at 256 bp resulted in amino acid change: Glu→Gly.

The single strand gFSHβ-CTP-α gene was successfully constructed by the combination of the C-terminal peptide (CTP) of carboxyl-terminal region of human chorionic gonadotropin with the goat FSH α-subunit and β-subunit genes. The gene was subcloned into expression vector pVITRO and transfected into Chinese Hamster Ovary (CHO) cells after gene sequencing. The results show that FSH analogous gene expression vector was successfully constructed , the CHO cells expressed gFSHβ-CTP-α were acquired and the quantity of expression is 0.120 mIU/mL. These results would lay the molecular foundation for studying the relation between structure and function of CTP and the production of longacting FSH.
Determination and Study on Cytb Gene Sequences of Alpaca

The sequences of cytochrome b were sequenced in the study. Homologous sequences were compared among llama, guanaco, vicuna, Bactrian camel, Arabian camel, and sheep. Base compositions and variance among these regions were calculated. Based on cytochrome b sequences the molecular phylogenetic trees were constructed by methods of Neighbor Joining and Maximum Likelihood, the taxonomic status of alpaca in the camelidae was discussed on the molecular level. The results showed that genetic difference between alpaca and vicuna, llama were comparatively more than genetic difference between alpaca and guanaco. The results made it possible that in the traditional taxonomy alpaca was breeding camel after domestic guanaco, and provided molecular basis for the traditional taxonomy.
SNP Detection and Its Association Analysis with Economic Traits in Exon 15 of Chicken Camk2d Gene

In this study, we cloned part DNA sequence of chicken Camk2d gene and found a C→T silent mutation at 51th bp in exon 15. Genotypes were detected by VspIPCRRFLP in 7 different native chicken lines in this test. The result of Chi square showed there had 3 genotypes (CC,CT,TT) in these 7 lines, and we could find the different distribution of three genotypes in different lines. According to the general linear model, we analyzed the correlation between gene and character among the three genotypes of Camk2d gene in 4 breeds (combinations). The result of variance analysis showed that there had significant correlation between this SNP site and abdominal fat (P<0.01), and correlation between this SNP site and pH value(P<0.05).

ApoB (apolipoprotein B), as the carrier of triglyceride, plays an essential role in the transport and metabolism of energy. So it was studied as one of the main candidate genes for growth and body composition traits. To discuss the relationship of apoB gene with growth and body composition traits especially for fatness in chicken, the eighth generation divergent lines of lean and fat broilers bred by Northeast Agricultural University were used in the present study. A SNP (T→G) in exon 26 of apoB gene, named T123G, was discovered by sequencing. Association analysis indicated that this SNP significantly related to body weight of 1, 3 week of age, abdominal fat weight and abdominal fat percentage at 7 week of age (P<0.05). Multicontrast analysis indicated that the birds with genotype GT were significantly higher than those with genotype GG for body weight at 1 week of age, the birds with genotype TT or GT were significantly higher than those with genotype GG for body weight at 3 week of age, and the birds with genotype TT were significantly higher than those with genotype GT or GG for abdominal fat weight and abdominal fat percentage. Population genetic analysis indicated that the frequencies of the three genotypes were significantly different in the two lines (P<0.05). So we putatively drew the conclusion that apoB or a tightly linked gene has important effects on body weight and deposition of abdominal fat in the chicken.
Cloning Partial Sequence of Myogenin in the Beijing Duck and Myogenin cDNA Expression Time Analysis

In our study, we used 4 pairs of primers to clone Beijing duck myogenin cDAN from breast muscle of 42day, 14day old Beijing duck and leg skeletal muscle of 0day old bird, 22day, 15day old hatched duck embryo. However, target gene was not amplified from the the reversetranscribed products. Data analysis, revealed that myogenin was expressed during the period of forming the myoblast cells. Myogenin expression may be found in cultures of muscle satellite cells or denervated animal skeletal muscle. 260 bp partial nucleotide sequence of myogenin exton 1 was amplified from DNA of metioned tissuesIt coded for a predicted protein of 86 amino acids. The predicted amino acid sequence has conserved bHLH domain and an overall similarity with turkey and chicken. The results provided a major base for researching myogenin expression, cloning complete cds of myogenin and polymorphisms analysis of duck myogenin gene.
Analysis of Genetic Diversity and Relationship among Local Chicken Breeds in Guizhou by Using Microsatellites Marker

The genetic diversities of 12 local chicken breeds in Guizhou and an introduced one were determined by seven microsatellite DNA markers, and the allele frequencies, gene heterozygosity (H), mean gene heterozygosity, polymorphism information content (PIC), mean polymorphism information content and genetic relationships of these breeds were also analyzed in this paper. The results illustrated that alleles and their frequencies in 7 loci marked by microsatellite showed differences among the 13 chicken breeds. Of these chicken breeds, the mean loci heterozygosity in the Yaoshan chicken is higher than the other ones, and that of the White Plymouth Rocks is the lowest. The mean PIC also showed that the genetic diversity in the Yaoshan chicken is the highest among these breeds. According to the clustering analysis, the 13 chicken breeds belong to 2 groups: one is the White Plymouth Rocks, the other group includes the 12 Local chicken breeds in Guizhou. And the 12 Local chicken breeds in Guizhou form 2 smaller groups. One group is composed of the Xingyi Native chicken, the Short Footed chicken, the Qiandongnan Xiao Xiang chicken and the Qiandongnan Xiao Xiang Silky chicken, and the other comprises the Long Footed chicken, the Yaoshan chicken, the Weining chicken,the Zhuxiang chicken, the Zhuxiang Silky chicken, the Black Wumeng Silky chicken, the Wumeng chicken, the Barred Wumeng Silky chicken.
Study on the hLF Gene Transgenic Goat Mammary Epithelial Cells

The recombinant plasmid containing human lactoferrin gene cDNA was transfected into goat mammary epithelial cells after liposome transinfection. Positive cells were selected by G418 and PCR. Positive cells were proliferated and induced by hormone. The result of Western blotting analysis on cultured cell supernatant showed that the transfected cells could express and secrete hLF protein, whose MW was 58 ku.
Study on Somatic Cell Clone and Conservation Technique of Luxi Cattle

In order to conserve our native excellent yellow cattle breeds, the somatic cell clone technique was studied, in which the bovine ear skin fibroblast in vitro cultured was used as nuclear donor. The fibroblast line of Luxi cattle (4 females and 1 male) for nuclear donor was established through tissue pieces culture method, and the oocytes of ovaries collected from slaughter house were cultured for nuclear transfer receptor. Experiment results indicate that the fusion rate and cleavage rate of reconstruction embryos were 62.5% (242/387) and 63.6%(154/242) respectively, and the development rate of blastocysts was 42.9%(66/154) after culture 7 days. The average cell number of inner cell mass and trophoblast was 37 and 47 respectively, in which 44.2% was inner cell mass. 10 blastocyts of reconstruction embryo were transferred to 10 recipient after cultured 7days in vitro, the pregnancy rate was 20%(2/10).

Leydig cells were isolated and purified by a discontinuous Percoll gradient (5%,30%,58%,70%).Purified cells were dispensed into 24-well plate and maintained for 24h in a CO2 incubator.Leydig cells were treated with taurine independently or coadministered with human chorionic gonadotropin(HCG),androstenedione,progesterone and cyclic adenosine monophosphate(cAMP).Testosterone levels were measured by radioimmunoassay. The results showed that taurine can affect testosterone secretion significantly in a biphasic pattern. Taurine at the concentration (0.1~100 μg/mL) increased testosterone secretion ,whereas the higher dose (400 μg/mL)inhibited testosterone secretion. Taurine can stimulate the convertion from androstenedione and progesterone to testosterone,which can also stimulate the testosterone secretion induced by HCG and cAMP.It is concluded that taurine can stimulate testosterone secretion ,which affect several aspects of testosterone biosynthesis.
Effects of Trehalose on Cryopreservation of Boar Semen

In order to improve boar sperm quality in frozenthawed process, trehalose was added into the basic extendersTrisCitric acidGlucose. The effects of trehalose on quality of cryopreservation of boar sperm were investigated. Moreover, in order to determine the optimum concentration of trehalose, we designed four different concentrations:25%,50%,75% and 100%(v/v), respectively. The results showed that the effects of the basic extenders supplemented with trehalose on cryoprotection of spermatozoa were better than that of traditional TrisCitric acidGlucose (TCG) without supplement. We found out that TCG extender with 25% trehalose concentration significantly enhanced motility, viability, mitochondrial activity, membrane integrity and acrosome integrity of boar sperm after freezing and thawing(P<0.05), and they were 41.38%,46.34%,44.56%,43.51% and 64.09%, respectively. Extender containing 25% trehalose could enable some improvement of cryopreservation of boar semen over the currently used TCG medium. Trehalose could decrease distinctly the percentage of spermatozoa capacitated caused by freezing and thawing(3.68%). Capacitation potential was improved significantly with the addition of trehalose in the diluents, especially at 25% concentration(41.82%). The optimal concentration of glycerol in extender was found to be 2%(v/v). Trehalose could improve spermatozoa quality after freezethaw process when there was glycerol in the extender. The results of present study indicated that there was a significant positive correlation among sperm motility, viability, mitochondrial activity, membrane integrity and acrosome integrity(P<0.01). However, the percentage of spermatozoa capacitated before capacitation treatment was highly negative correlated with sperm quality index in this study(P<0.05).

Effect of Cyclic High Temperature on Mitochondrial ROS Production, Ca2+ATPase Activity and Breast Meat Quality of Broilers

The present study was conducted to determine the adverse effects of cyclic high temperature on mitochondrial function and breast meat quality of broilers. One hundred eighty 4-week-old broilers were allocated to three treatments according to following design: 25 ℃, ad lib. feeding (Control); 25 ℃, pairfeeding with the high cyclic group (PF); and 28 ℃/34 ℃ daily cyclic high temperature, ad lib. feeding (Cyclic). Broilers were kept in three environment control chambers for three weeks. Heat exposure significantly elevated hepatic mitochondrial H2O2 production and muscular and hepatic lipid peroxidation, significantly inhibited muscular and hepatic mitochondrial Ca2+-ATPase activity. Increased plasma CK and LDH were found under cyclic high ambient temperature, which indicated that the muscle intracellular free Ca2+ was increased and muscle cell membrane integrity was damaged. At 49 day, Consistent with cyclic high temperature induced alterations in mitochondrial function were changes in breast muscle glycolytic metabolism as indicated by lower muscle pH immediately postslaughter (pHi), and higher drip loss, L*and shear force. At 25 ℃, restrict feeding had no significant effects on mitochondrial function, muscle pH, L*, and drip loss, which indicated that effects of cyclic high temperature on mitochondrial function and breast meat quality of broilers were not associated with declined in feed intake.
Effects of Neuroendocrine Factors on mRNA Abundance of HSL and LP in Neonatal Calf Adipocytes in Vitro

To clarify regulation of INS, GLN and LP on the mRNA expression of HSL and LP in vitro neonatal calf adipocytes, different concentrations of INS (0, 5, 10, 20, 50 and 100 mmol/L), GLN (0, 25, 100, 200, 500 and 1 000pg/mL),and LP (0, 25, 5, 10, 50 and 100 ng/mL) were added respectively into media of primary monolayer culture calf adipocytes. After 12 h cultured, mRNA abundance of HSL and LP in the adipocytes was detected by FQPCR respectively. The results showed that INS plays a dosedependently inhibitory role in abundance of HSL mRNA and a stimulating role in LP mRNA in calf adipocytes in vitro. However, action of GLN was contrary to INS in abundance of HSL mRNA, and plays a no role in abundance of LP mRNA in calf adipocytes in vitro. With concentrations of LP, abundance of HSL mRNA in calf adipocytes in vitro first dosedependently increased, then decreased, but abundance of LP mRNA in calf adipocytes dosedependently decreased in vitro.

Comparative Studies on Immune Responses of IBV DNA Cationic Liposome and Chitosan Complex Vaccine

Liposome/DNA and chitosan/DNA complexes of infetious bronchitis virus which were prepared were immunized health chickens via intramuscular injection(i.m.)and eye drop, intranasal administration (i.n), with naked DNA vaccine as control. Fluorescence activated cel1 sorter(FACS) and indirect ELISA methods were used to detect the number of CD4+ and CD8+ T lymphocytes and antiIBV antibody IgG.All the chickens were challenged with 0.2 mL of 103EID50 IB SAIBK on day 35 after vaccination. Result: The number of CD4+ T lymphocyte of the experiment groups at 1-3 weeks post inoculation was higher than that of the control group (P<0.01 or P<0.05),and between the experiment groups, the number of CD4+ cell of liposome/DNA vaccine 10 days after inoculation was higher than that of chitosan/DNA vaccine via i.m, but only appeared significant difference at the 15th day, but it was opposite by i.n.administration. The number of CD8+ T lymphocyte of the experiment groups were higher than that of control group(P<0.01 or P<0.05)on day 15 after vaccination, however it was not different between the experiment groups. The level of specific IgG of all groups only appeared insignificant differece. The livability of chickens of liposome DNA group(i.m.)was 100%,other experiment groups had 4% or 8% mortality.The mortality of all experiment groups was lower that of control group(12%).From these results we can draw the conclusion that liposome and chitosan were good adjuvant or vector of DNA vaccine, and the immune responses of liposome/DNA vaccine was better than chitosan/DNA vaccine via i.m.
Development of Monoclonal Antibodies against VP3 of IBDV and Identification of Antigenic Epitopes

Female, 6-8 weeks old BALB/c mice were immunized with the purified recombinant fusion VP3 of IBDV. After three immunizations, the spleen cells from immunized BALB/c mice were fused with mouse myeloma cells SP2/0. By 3 cycles of limited dilutions, we obtained four strains of hybridoma cells, named HRB7B,HAR-7C,HRB-3F and HRB-10E. Isomerism and stability analysis indicated that those monoclonal antibodies had specific reaction with the VP3 of IBDV. The titer of four antibodies were up to 5.0×102 in cell culture supernatants and 6.4×106 in ascetic fluids. Plusing ELISA showed that in four cell strains, HRB-7C and HRB-10E have the similar antigen epitopes. Moreover, the study identified the antigenic epitopes by peptide scanning, further structural and function analysis of IBDV. This work could provide the basis for the development of immunitybased prophylactic, therapeutic and diagnostic techniques for the control of IBD and further structural and function analysis of IBDV.
An Indirect ELISA for Detection of Antibody against Infectious Bovine Rhinotracheitis Virus Using Recombinant Truncated Glycoprotein B Expressed in E.coli

The gene, which codes the amid acid residency from 192th to 266th of Infectious Bovine Rhinotracheitis Virus(IBRV) glycoprotein B(gB), was amplified by PCR technique with specific primers after analyzing glycoprotein B with biosoftware DNAstar. Then the amplified product was cloned into Tvector. After identifying with doubleenzyme cut and sequencing for the accuracy of the product of amplification, the expected band on the agarose gals was recovered and directional inserted into pET32a vector. The recombinant vector was transformed into BL21. SDS-PAGE analysis showed that the fusion protein of 29.2 ku as expected was in solution forms partially. The recombinant protein was purified by affinity chromatography, the concentration of purified protein was 2.000 mg/mL and the purity was 79.2%. The recombinant protein was checked by the indirect enzymelink assay, which showed that it retains antigenicity of the nature protein. An indirect enzyme linkedassay(antigen concentration: 50 μg/mL; serum dilution: 1∶40) was developed with the recombinant protein as a diagnosis antigen, the positive criterion was S/P>0.395There was no cross reaction with positive sura of other six diseases, including BEF(Bovine ephemeral fever), CBPP(Contagious bovine pleuropneumonia), BVD-MD(Bovine viral diarrheamucosal disease), BTB(Bovine tuberculosis), BPB(Bovine paratuberculosis) and BAD(Bovine Akabane disese). Comparing with the Virus Neutralization testing, the speciality was 83.3%; the sensitivity was 90.9% ;the accordance rate was 88.9%. Comparing with the IBR ELISA kit from France, the speciality was 92%; the sensitivity was 92.7% ; the accordance rate was 92.5%. The assay was confirmed to be specific and sensitive by the results above, suggesting a good application prospect.
Phylogenetic Analysis of the Surface Glycoprotein Genes of Three H4 Avian Influenza Virus Isolates from Water Fowl

Several H4 subtype avian A influenza viruses were isolated from waterfowls in some live bird markets when we surveying the ecology of the influenza in East China for more than four years and identified by specific RTPCR. In this paper, the hemagglutinin (HA) gene and neuraminidase (NA) gene of three representative virus named A/Duck/Yangzhou/216/2002 (Dk/YZ/216/02), A/Duck/Yangzhou/526/2003(Dk/YZ/526/03)and A/Duck/Yangzhou/36/2004(Dk/YZ/36/04) had been sequenced and their pathogenicity to SPF chickens had been determined. The results showed that the HA nucleotide sequence of Dk/YZ/216/02 have high identity with Tk/Minnesota/833/80(H4N2),that of Dk/YZ/526/03 and Dk/YZ/Z36/04 have high identity with Budgerigar/Hokkaido/1/77(H4N6); Phylogenetic analysis of the N6 NA genes revealed that the NA gene of Dk/YZ/Z36/04 had genetically close relationships with that of H4N6 viruses isolated from pigeon in jiangxi China, the NA gene of Dk/YZ/526/03 had genetically close relationships with that of H9N2 viruses isolated from duck in Hokkaido. The sequences of cleavage site between HA1 and HA2 consist of P-E-K-A-S-R in all three AIVs, coincide with that of identical low pathogenic AIV, and none of the chickens inoculated with the three AIVs displayed any obvious signs of clinical disease.
The Investigation on the Molecular Epizootiology of Newcastle Disease Virus Isolated from China in 2005

Eighty-three strains of Newcastle disease virus(NDV) were isolated from cases during epidemic in different parts of China in 2005. The main functional region of the F gene were amplified by RT-PCR and sequenced. All the sequences obtained from the NDV isolates were submitted to GenBank, the accession numbers were DQ439866-DQ439948.The sequence analysis showed that the length of the F gene segments amplified were 535 bp. For genotyping, phylogenetic tree based on the nucleotide 47-435 of the F gene were performed using sequences of 83 isolates and other NDV sequences obtained from GenBank. The results revealed that all the isolates belonged to 5 genotypes(Ⅶ,Ⅵ,Ⅲ,Ⅱ and Ⅰ). Most of the isolates (65 strains) were classified into genotype Ⅶ. There are 9 strains belonged to genotype Ⅱ, which were similar to the vaccine strain LaSota. Five strains were proved to be genetype Ⅰ, which were similar to V4. Three strains were genotype Ⅵ, and one isolate, NDV05-041, belonged to genotype Ⅲ. It indicated that the genotype Ⅶ NDV was the major pathogeny causing the epizootic of ND in 2005 in China.

Effects of Gushukang on Bone Metabolism of Laying Hens

Effects of Gushukang(GSK)on bone metabolism of laying hens were studied. 90 layers of 58-week old were randomly assigned to three groups. The control was provided with basal diet, the GSK group was provided with basal diet with addition of 1% GSK, the ipriflavone(IP)group was provided with addition 25 mg/kg IP. The experiments lasted for 90 days. The results indicated that the levels of Ca, P, AKP, TRACP, BGP in the serum from GSK and IP groups were decreased significantly; but bone index, bone radiographic density, cortical width, cortical area and cortical area ration of humerus, tibia and femur, and the level of serum IGF1 were higher significantly in the GSKgroup and IP group than the control. There was not significant difference between GSKgroup and IP-group. It was revealed that GSK can accelerate bone formation, inhibit bone resorption and improve bone metabolic profile of layers during the later laying period.

Effect of Zinc and Selenium Level in Diet on the IgA+cell in the Gut of Broiler

To explore the effect of trace elements zinc, selenium and their interaction on the gut barrier function in the broiler, twenty-four one-day-old AA broilers were divided into three groups, and were fed with the different diets including high zinc-selenium (Zn 1 000 mg/kg, Se 5 mg/kg), low zinc-selenium (Zn 34 mg/kg, Se 0.08 mg/kg), and normal zincselenium (Zn 50 mg/kg, Se 0.15 mg/kg), respectively. After fortyfive days, the broilers were killed and the distribution and quantity of IgA+-cell and IgA positive reaction substance in the gut were observed by immunohistochemistry. The results showed that the IgA+cell and IgA positive reaction substance were located in the tip of villus, lamina propria, surround intestinal gland, and lymphatic nodule of cecum. In two groups of high zinc-selenium and low zincselenium, the quantity of IgA+-cell and IgA positive reaction substances were decreased(P<0.01), especially in the high zincselenium group. The distribution and the number of the IgA+-cell and IgA positive reaction substances were normal in the normal zinc-selenium group. This study indicated that the low zincselenium or high zinc-selenium would repress production of IgA, and the supplementary level of selenium at 0.15 mg/kg and zinc at 50 mg/kg in diet should be an appropriate ratio which would keep the normal gut barrier function in the broiler.

The effect of a traditional chinese medicine preparation(TCMP) on the change of enzyme activities, such as serum transaminase, hepatic antioxidant enzyme in eel(Anguilla anguilla) was evaluated in this study. Six eels, acting as Cupoisoned control fish, were exposed to waterborne Cu(0.12 mg/L) in moderately hard water for 4 days and kept at background Cu levels(0.004 mg/L) in the following 4 days. Eighteen eels were disposed in the same way as the Cu- poisoned control fish in the first 4 days, and then six eels among them were dipped in 200 mg/L water solution of Baogan Jiedu decoction(BJD), which was made up of Yinchen, Longdan, Gancao, Dahuang and Zhizi, in the following 4 days as low-dosage-drug-treated fish,other six eels were dipped in 400 mg/L BJD as high-dosage-drug-treated fish, and the remnant six eels were dipped in 100 mg/L of Jiangmeiling capsule containing Wuweizi as drugtreated control fish. Six eels as control fish were kept at background Cu levels(0.004 mg/L) in the whole trial course of 8 days without any disposition. The fish behavior and outer symptom were observed during the trial period. At the end, the fish were killed, and their blood were immediately collected and centrifuged to collect serum for testing the activity of its transaminase(AST and ALT).Subsequently, liver was dissected out, rinsed with doubledistilled water. A fraction of the liver was treated for the assaying of antioxidant enzymes(CAT,SOD and GSHPx). The results indicated that, compared with the control fish, the levels of ALT and AST were elevated significantly(P<0.01), the activities of CAT and GSHPx decreased sharply(P<0.01), and the activity of SOD was activated slightly(P<0.05) in Cu-poisoned control fish. And in drugtreated fish and drugtreated control fish, the activities of ALT and AST droped obviously(P<0.01), the activities of CAT and GSH-Px increased remarkably(P<0.01), the activities of SOD were enhanced further more than the Cu-poisoned control fish. Overall, these results as mentioned above demonstrated that the Baogan Jiedu decoction can enhance the activities of ALT and AST in the serum, and also bring the activities of CAT, GSHPx and SOD going up significantly. As a result, the preparation is suggested a effective liverprotective medicine against the liver injury induced by poison.
The Innervation and Location of the Primary Sensory Neurons on the Hens’ Uterus of Uterine Tube by CB-HRP Methods

The location and distribution of the primary sensory neurons on the hens’ uterus of uterine tube were determined by retrograde trace method.CBHRP solution was injected into the wall of the uterus of ten Issabrown hens. The results showed that the primary sensory neurons of Issabrownhens uterus of uterine tube were innervated by bilateral spinal ganglia from T1 to LS13,bilateral jugular and nodose ganglia.The numbers of labeled neurons in the left were more than those in the right,and that in the spinal were more than those in the jugular and nodose ganglia. A lot of labeled neurons in the spinal ganglia were accumulated on the segments T5-LS1 and LS8-LS11.The greatest concentration of labeled neurons were in T7 and LS11.It was revealed that hens’ uterus of uterine tube were innervated by bilateral spinal and vagus afferent nerves,in which mainly conducted by ipsilateral or left side spinal and vagus nerves,and the spinal was dominat,although the uterus of uterine tube of hens was the unilateral visceral organ.

Effects of in vitro Culture Systems on the Bovine Embryo’s Sex Ratio

The experiment was carried out to study the effects of in vitro culture systems on the bovine embryo's sex ratio. The results indicated that the sex ratio of male blastocysts versus female blastocysts was different from the expected 1∶1 ratio but no significant different (P>0.05) in the two culture systems supplemented with FCS. While the total sex ratio of blastocysts derived from all kinds of culture systems was different from the 1∶1 ratio significantly (P<0.05). It demonstrated that the capacity developing to the blastocyst stage of male embryos were stronger than that of female embryos in vitro culture system, and the presence of serum has a beneficial effect on the development of male embryos.
Influence of Different Electricfusion Conditions on the Development of ransgenic Nuclear Transfer Embryos of Saanen Dairy Goat

In this study using the transgenic (GFP) fibroblasts of Saanen dairy goat as donor cells and using the abattoirderived oocytes of normal goat as recipient cells the nuclear transfer experiments had been processed. During experiments the optimization of electricfusion experiment parameters of transgenic nuclear transfer embryos had been done. For the treatment of transgenic reconstruction embryos of Saanen dairy goat, the electricfusion parameter of group B in experiment plans (1.5kV/cm for 30 μs and pulse interval for 1s) is most suitable for the transgenic reconstruction embryos of Saanen dairy goat, the electricfusion rate of 89.4% for cell-cytoplast complexes and the cleavage rate of 68.0% for reconstructed embryos have been obtained.
Preparation of Colloidal Gold Strip for Semiquantitative Test of Antibody against Newcastle Disease Virus

To develop a “semi-quantitative RUN test” method to detect antibody against Newcastle disease virus, Tri-sodium citrate with aqueous gold chloride were warmed up and mixed to make gold sol. The 20nm-diameter colloidal gold was used to mark NDV antigen and the colloidal gold strip was prepared for semiquantitative test of antibody against NDV. 60 samples were tested by the colloidal gold strip. The coincidence between the colloidal gold strip and HI was 93.8%. The least test level of the strip was 4log2 detected by HI experiment, and it didn't exist crossreaction with other serums against corresponding virus . The strip was still effectual after storing three months at room temperature . Colloidal gold kit is a rapid method in clinic practice and could be used in surveillance of antibody against NDV.
