In this research, we have mapped 6 QTL for 11 meat quality traits, including intramuscular fat of m. Longissimus dorsi (IMF), water moisture in m Longissimus dorsi (WM), water holding capacity of m. Longissimus dorsi (WHC), drip loss rate of m. Longissimus dorsi (DLR), meat pH of m. Longissimus dorsi, (pH1), meat pH of m. Biceps femoris (pH2), meat pH of m. Semispinalis capitis (pH3), meat color value of m. Longissimus dorsi (MCV1), meat color value of m. Biceps femoris (MCV2), meat marbling score of m. Longissimus dorsi (MMS1) and meat marbling score of m. Biceps femoris (MMS2), in a 214 pigs resource population including 180 F2 individuals, using 23 microsatellite marker loci on SSC (Sus scrofa chromosome) 4,8 and 13. The results indicated that two highly significant QTL (P≤0.01) for IMF (at 55 cM on SSC13) and WM (at 10 cM on SSC4) were detected, and 4 significant QTL (P≤0.05) for MCV2 (at 110 cM on SSC8 and at 70 cM on SSC13), MMS1 (at 185 cM on SSC8) and MMS2 (at 185 cM on SSC8) were mapped. The phenotypic variances for which these QTL have accounted were range from 0.16% (QTL for MCV2) to 17.84% (QTL for MMS2).

Polymorphisms in exon 14 of Mx1 gene were detected by PCRRFLP in Sutai pigs.Six genotypes and three alleles were observed when exon 14 of Mx1 gene was digested by Hin6Ⅰrestriction enzyme. The results of Chi Square test indicated that the frequencies patterns of exon 14 of Mx1 gene were fit with Hardy-Weinberg equilibrium in Sutai pigs population. Correlation between genotypes and reproductive performances (total litter size, number born alive, birth litter weight, number weaning piglets and weaning litter weight in the third litter) of Sutai pigs was similarly analyzed. The results indicated that reproductive performances of Sutai pigs with BC genotype were significantly (P<0.05)higher than those with BB, AA, AB, AC and CC genotypes. Three new points mutation were identified by sequencing PCR products, two of which resulted in Thr→Ala and Glu→Arg were missense mutation, and the last was a silence mutation. Glu→Arg point mutation and silence mutation were appeared in the individuals with BB genotype. The results also indicated that the point mutation Glu→Arg may influence the virus resistance of Mx protein in pigs

Three SNPs loci in IGF-Ⅰ gene in 180 Wenling Grass-chicken including 3 lines were detected by PCR-RFLP method,and its genetic effects on carcass traits were also analyzed. The mutations of T→C,C→A and C→T were identified by PstⅠ,HinfⅠand TaqⅠ, respectively. Three genotypes were acquired in each locus,and line A was at the Hardy-Weinberg equilibrium (Ρ>0.05) in each locus. The analysis of variance showed that the genotypes of each locus had significant or suggestive difference (Ρ≤0.01 or 0.01<Ρ≤0.05) in partial carcass traits. The multiple comparisons indicated the Least Squares Means of 3 genotypes in each locus had the property of mutant genotype>heterozygous genotype>wild genotype in 6 carcass traits,and the com-genotypes QF∕QF and QE∕QF were suggestive higher (0.01<Ρ≤0.05) than PE∕QE in 4 carcass traits. And the mutant allele of each locus had additive effect on partial carcass traits. The genetic distance between line A and B was 0.005 3,indicating they were in the same branch through cluster analysis.

In present study,8 populations (including 5 pure lines and 3 crossbreeds developed by Dahen Poultry Breeding Company and Sichuan Animal Science Academy) were used to analyze MC3R gene polymorphism by PCRSSCP.Result indicated that there was one mutation sites in 8 populations (A 1424 G). The statistics analysis results showed that the MC3R polymorphism was no significant difference for SEP and LMP trait (P>0.05), while there were significant difference (P<0.05) and high significant difference (P<0.01) for other slaughter traits. In addition, the significant difference of CP and IMF value were also observed only at meat quality traits (P<0.05).

CD154 plays an important role in the interaction between antigen-specific T lymphocytes and antigen-presenting cells. In this study, the full-length cDNA encoding chicken CD154 was amplified from isolated peripheral blood mononuclear cells (PBMC) of Sanhuang chicken using RT-PCR- Sequence analysis showed that the chicken CD154 is comprised of 819 base pairs in length and encodes a 272 amino acid (aa) protein.The full-length cDNA fragment of chicken CD154 was further subcloned into prokaryotic expression vector pET-28a, resulting in the expression plasmid pET-28a-CD154. However, no specific expressed protein was detected after transformation of BL21(DE3). The truncated fragment of chicken CD154 (CD154d) without signal peptide (1-22 aa) and transmembrance region (23-46 aa) was cloned using PCR and further subcloned into downstream of hexahistidine sequence of pQE80. Expression of a fusion protein (6×His-CD154d) with 27 ku molecular mass was observed in E. coli DH5α, and the result of Westernblot demonstrated that the fusion protein can be recognized by anti-6×His monoclonal antibody. In addition, the fulllength cDNA fragment of chicken CD154 was inserted into downstream of EGFP gene of eurokaryotic expression vector pEGFP-C1, resulting in the expression plasmid pEGFP-C1-CD154. After transfection in Hela cells, the fluorescence of the fusion protein EGFP-CD154 was observed on cell membrane by fluorescent microscope, indicating that the chicken CD154 is a membrane-associated protein. The present study laid a foundation for further investigating the structure and function of chicken CD154 and its adjuvant potency in chicken vaccines.

According to the real breeding and production condition for beef line of Luxi in nucleus and commercial, Profit function were constructed on the basis of two-tie open nucleus system with the fixed herd-size, which consisted of 6 animal groups that were calves, heifer, young bull, cow, bull and fatten cattle, including 9 objective traits which were weaning weights, yearling weights, mature cow weight, fatten daily gain, carcass grade, dressing percentage, age of first calving, calving interval and productive lifetime. The profit was estimated for current system and economic values were derived from profit function by partial budgeting method for 9 objective traits. Discounting genetic coefficients were calculated and economic values were standardized by genetic standard deviation to obtain economic weights and relative economic weights of objective traits. The results show that total profit of 6 animal groups were -1 033.95 yuan, and economic values for 9 objective traits were 1.44 yuan/kg, 1.49 yuan/kg, -3.02 yuan/kg, 1.81yuan/g, 315.45 yuan/class, 66.85 yuan /%, -0.54 yuan/d, -4.43 yuan/d, 0.09 yuan/d; and these of relative economic weights were 0.11, 0.11, 0.14, 0.26, 0.14, 0.12, 0.01, 0.08, 0.03, respectively. The results of this study suggest that growth traits should be mainly strengthened to select from beef and labor dur-purpose to special beef direction, and that attention should be give to carcass and reproduction traits simultaneity.

Nine wethers of Inner Mongolian White Cashmere (IMWC) goats (25-30 kg) with the same father, fitted with permanent cannulas at the proximal duodenum, and with two sides temporary carotid venous catheters were used to investigate the effect of absorbable amino acid pattern in the small intestine, i.e. M, M90+C10 and M80+C20, on FSR of proteins in the jejunum, liver, muscle and skin tissues in IMWC goats. The results showed that absorbable amino acid pattern had significant effect on protein FSR in skin tissue, and M90+C10 and M80+C20 were significantly greater than M (P<0.01). There were no significant effect of amino acid pattern on the FSR in jejunum, liver and muscle (P>0.05). It also was showed that absorbable amino acid pattern had significant effect on distribution of amino acids between skin and muscle(P<0.05).

To investigate the effect of fibrolytic enzymes on gastrointestinal morphological structure in kids, thirty healthy Boer male kids were chosen at similar birth weight to divide into two groups, the control and the treatment group (adding 0.2% fibrolytic enzymes),and were weaned at 80-day old. Alfalfa hay were provided at 20-day old. Three kids chosen from each group were slaughtered at 2-, 3- and 4-month old, respectively. The results indicated that the length(P < 0.01), width(P > 0.05) of rumjen papilla and surface area of rumen papilla per cm2(P < 0.01) were increased in the treatment group at the age of 3 and 4month, respectively, compared with control group, however the number of rumen papilla per cm2 was decreased (P < 0.01). The fibrolytic enzymes increased significantly the villus length of duodenum at 3month age (P < 0.05), and tended to increase the recess depth and mucous coat depth at the age of 3 and 4 month(P > 0.05).

The experiment was conducted to investigate the effect of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on peripheral blood lymphocyte proliferation and intracellular signaling molecules in weanling pigs. The results showed that EPA and DHA inhibited lymphocyte proliferation (P < 0.05) and stimulated nitric oxide (NO) (P < 0.001), cAMP (P < 0.001) and cGMP (P < 0.001) production and cAMP/ cGMP ratio in a dose-dependent manner. EPA and DHA induced a momentary rise of intracellular Ca2+, and the rising extent was dependent on the concentration of EPA and DHA. EPA and DHA also inhibited protein kinase C activity in a dose-dependent manner (P < 0.05), and had no effect on protein kinase A activity. These results suggested that EPA and DHA exert an inhibitory effect on lymphocyte function, possibly due to alteration of intracellular signal transduction.

In order to study the effect of cell mediated immunity regulation of duck IFN-α eukaryon expression plasmid (pcDNA-SDIFN-α) on DPV attenuated vaccine in ducks , pcDNA-SDIFN-α was administrated to 28-day old ducks at the dose of 1, 3, 6 μg/duck, respectively, by gene-gun. PBS and empty vector pcDNA were used as control.15 days later, all ducks were injected with DPV attenuated vaccine, and blood samples were collected at 3, 7, 14, 21, 28, 35, 49, 63, 84 days after injection. T lymphocyte proliferation tests (MTT) were used to detect the T-lymphocyte proliferation in peripheral blood(PBL) of ducks. Blood samples collected at 7, 14, 21, 28, 35, 49 days were detected by fluorescence activated cell sorter (FACS) for recording the number of CD+3 T lymphocytes of ducks Results were as follows: ① Reaction of T lymphocytes in PBL to ConA (OD value)of ducks treated with pcDNA-SDIFN-α were higher than that of PBS and pcDNA control groups in 3—84 days. There were highly significant difference between 1 μg/duck group and two control groups in 3—84 days (P≤0.01), between 3 μg/duck group and two control groups in 3-84 days (P≤0.01,P≤0.05), and between 6 μg/duck group and two control groups in 7-49 days (P≤0.01,P≤0.05).The significant difference was also present between 1 μg/duck and 3,6 μg/duck groups in 3-35 days(P≤0.05). Moreover, there was no significant difference between 3 μg/duck group and 6 μg/duck group (P≥0.05). pcDNA control group was higher than PBS control group, but no difference was found (P≥0.05).②Change of the number of CD+3 T-lymphocyte of ducks administrated with different doses of pcDNA-SDIFN-α were higher than that of PBS and pcDNA control groups in 7-49 days.1 μg/duck group was significantly higher than PBS and pcDNA control groups in 14-49 days (P≤0.01), there were significant difference between 3 μg/duck group and two control groups in 21-49 days (P≤0.01,P≤0.05),significant difference between 6 μg/duck group and two control groups in 7-49 days (P≤0.01,P≤0.05). No significant difference was found among the groups of 1, 3, 6 μg/duck (P≥0.05).There was no significantly difference between two control groups(P≥0.05). The results indicate that pcDNA-SDIFN-α administrated 15 days before injection of DPV attenuated vaccine could significantly enhance cellular immunity induced by DPV attenuated vaccine. pcDNA-SDIFN-α is an excellent DPV attenuated vaccine molecular adjuvant, best result could be obtained with the dose of 1 μg/duck of pcDNA-SDIFN-α inoculated by gene-gun.

The recombinant expression vector pGEX-IL-4 was induced by IPTG in optimal conditions in E.coli.SDS-PAGE analysis showed that the expressed fusion protein with a molecular weight of 38 ku existed in the inclusion body. The fusion protein, which was of 25% in total bacterial proteins, was denatured, renatured, and purified with saturated ammonium sulfate precipitation and Sephadex-G100 with the purity of above 90%. MTT analysis indicated that purified protein has good bioactivity, which has laid a foundation for the production and development of recombinant protein of porcine IL-4.

Four-week old SPF chickens were inoculated with fowlpox virus vaccine coexpressing gene S1 of infectious bronchitis virus (IBV) and the chicken interferon-γ gene(rFPV-IFNγS1). SPF chickens were challenged with a heterologous IBV strain LSC99I to investigate protective efficacy of the recombinant virus to heterologous virus after three weeks postinoculation The results showed that chickens immunized with rFPV-IFNγS1 developed IBV specific antibodies in serum one week after immunization, the ratio of CD4+ and CD8+ T lymphocyte in chicken’s peripheral blood of immunized group was a little higher than that of the nonvaccinated group, but the difference was inapparent.Morbidity and mortality of immunized group were 50% (5/10) and 20% respectively after challenge with LSC99I compared with 100% (10/10) and 50% (5/10) of non-vaccinated group,chickens in immunized group had slighter pathological changes when compared to that of non-vaccinated group, but the difference was indistinctive. The length and titer of virus shedding for two groups showed no significant difference. Bodyweight analysis indicated that chickens in both groups showed inapparent weight change before challenge; after challenge, chickens in immunized group had a higher weight gain than chickens in non-vaccinated group. In conclusion, rFPV-IFNγS1 cannot protect chickens against the challenge of heterologous IBV strain LSC99I although it can produce IBV-specific antibodies in chickens, chicken IFN-γ is well expressed in vivo and play a role in immunomodulatory which can alleviate the effect of recombinant virus immunization on chicken bodyweight gain.

A nested polymerase chain reaction (N-PCR) assay for direct detection of toxigenic Pasteurella multocida( T+ Pm) in nasal swab specimens collected from pigs was developed using 2 pairs of primers that were designed according to the published sequences of T+ Pm toxA, The size of amplicons were 864 and 447 bp, respectively. The assay could detect 26 CFU microorganisms. Mean while, no false amplification in detection of Escherichia coli, Streptococcus, Staphylococcus, Bordetella bronchiseptica, Nontoxigenic Pasteurella multocida, Haemophilus parasuis and Actinobacillus pleuopheumoniae could be observed. The nested PCR and bacterial isolation were employed simultaneously to examine 146 nasal swabs collected from 5 herds known to be infected with toxigenic Pasteurella multocida, 44 and 97 specimens were positive and negative for both isolation and nested PCR, respectively, 5 were positive for nested PCR only. The coincidence between bacterial isolation and nested PCR is 96.58%. These results indicated that the developed nested PCR might be a promising approach in clinical screening and detection of toxigenic Pasteurella multocida in a large number of swine swabs.

The Sulfonamides-resistant genes Sul1 ,Sul2, Sul3 were detected by duplex PCR from 33 Salmonella strains and 103 E.coli strains from pig and chicken The results showed that Sul1,Sul2 genes of 17 Salmonella strains were detected from 19 SD resistant Salmonella strains demonstrated by MIC, and the detection rate was 89.47%. The coincidence rate between duplex PCR assays and MIC assays was 89.47%. Sul1 ,Sul2 genes of 53 E.coli strains were detected from 56 SD resistant E.coli strains. The rate of coincidence of duplex PCR assays and MIC assays was 94.64%.The rate of coincidence between duplex PCR assays and common PCR assays is 98.5%. It was only taken 7 hours to complete the whole duplex PCR test.

A simple and rapid method for purifying merozoites of Theileria spp. from infected erythrocytes in sheep or goats was developed. Infected erythrocytes were effectively lysed using hemolysin Ah-1 produced by Aeromonas hydrophila, and the lysate was subjected to centrifugation in a Percoll discontinuous density gradient at common speed. Light microscopical study showed that the merozoites purified retained their intact basic structure and were free of infected erythrocyte membrane and other host-cell components

Spectrophotometer and nitric oxide synthase(NOS) detection kit were used for finding out law of NOS activity in various brain regions of different agegroups rabbits.The results showed: ①NOS activity was detected in every area of rabbit brain, furthermore NOS activity in rabbit cerebellum was higher than diencephalon and brain stem, the lowest activity was found in cerebral cortex②Basically, NOS activity of the young rabbit had developed to the adult rabbit’s level.It developed from maturation to senescence gradually with age.To the aged rabbit, the NOS activity appeared senescence obviously. NOS activity was higher in the adult group than that of young group, juvenile group and the youth group. The lowest NOS activity was found in the aged group. The results indicated that the decreased NOS activity and diminished NO production might weaken role of No on nerve regulation with the rabbit’s doating, which perhaps effect the senescence of rabbit brain, arterial sclerosis and increase in blood pressure.

The dorsal root ganglia (DRG) of goat fetus formed before 6th embryonic week, but the morphology of neurons inside ganglia were not very distinct until 8th embryonic week. The increase of ganglion volume mainly lied in growth of neurons and increasing of nerve fibers and neuroglia cells, but not lied in marked increase of neurons in number. Neurons could be divided into dark subpopulation and bright subpopulation according to their shape and staining extent, or three subpopulation—large size, medial size and small size according to cellular diameter. NGF and TrkA immunoreactivity were present in DRG of goat fetus. NGF-IR mainly localized in cytoplasm of three subpopulation, especially after 14th embryonic week; TrkA-IR occurred after 10th embryonic week, and were mainly distributed in cytoplasm and nucleus of medial-sized and smallsized neurons. No TrkA-IR could be detected on large-sized neurons, but TrkA immunostaining distributed in satellite cells surronding large-sized neurons The results suggested that NGF play an important role both in regulating the physiological function of developing sensory neurons derived from peripheral neural crest and in growth,development and maturation of satellite cells.

The objective of this study was to investigate the changes of the number of lymphocyte, goblet cell reside within the intestinal musocal epithelium and mast cells’ density within intestinal wall area in beef calf.Histochemical and the Scion image software analysis techniques were used. Data were collected from duodenum, jejunum and ileum of 4 beef Limousin crossbred calves at 1-, 4- and 6-month of age, respectively.Results showed that intraepithelium lymphocyte number and mast cells’ density gradually decreased, but goblet cell number gradually increased from duodenum, jejunum to ileum in beef calf at same month of age.Intraepithelium lymphocyte number of duodenum, jejunum and ileum in beef calf, which significantly differed (P<0.01)each other at 1- and 6-month of age, intraepithelium lymphocyte number of duodenum at 4-month of age was the mostGoblet cell number of duodenum, jejunum and ileum in beef calf was significantly different (P<0.01)each other at 1-month of age.At 4- and 6-month of age, goblet cell number didn’t differ significantly(P>0.05) between duodenum and jejunum, but the numbers of these two segments were less (P<0.01)than that of ileum.As for mast cells’ density within intestinal wall area of duodenum, jejunum and ileum in beef calf,they significantly differed (P<0.01)each other at 1-, 4- and 6-month of age, respectively, mast cells’ density within intestinal wall area was least at 4month of age.Our results indicated immune responses may involve the coordination and potentially redundant development of calf.

The polymorphism of the exon 2 of MHC-DRB1 was investigated by nested PCR-RFLP technique at three loci digested with SacⅠ,Hin1Ⅰand HaeⅢ,respectively, in a total number of 211 individuals of Chinese Merino(Xinjiang Army Wasteland Reclamation) sheep, and genotypic frequencies and allelic frequencies were counted. The results showed that the extensive polymorphisms of the exon 2 of MHC-DRB1 were found at three loci digested with SacⅠ,Hin1Ⅰ and HaeⅢ,respectively, in Chinese Merino sheep, these sites were controlled by two, two and six alleles, respectively. As some base mutations were found in MHCDRB1 locus, and according to the results in this paper, twentyfour alleles were found in Chinese Merino(Xinjiang Army Wasteland Reclamation) sheep. The results of χ2 test showed that genotypes of MHC-DRB1 digested with SacⅠand Hin1Ⅰexcept HaeⅢ(P<0.01)fitted with Hardy-Weinberg equilibrium(P>0.05)

The effect supplementing with Chinese Medicine effective constituents Baicalin(Bai)and Ligustrazine(Lig) into culture medium on 2-cell mouse embryos in vitro culture was compared in this study. And the morula and blastocyst in vitro culture were conserved through two freezing process The frozenthawed modality choiceness embryos were cultured to blastocyst by 8-14 h or 6-8 h and transferred to recipient uterus. The results showed that the hatching rate of embryos at 120 h from Bai(80.6%) and Lig(78.3% ) were significant higher than control group (51.8%), respectively (P<0.01)According to the cell amount of embryos hatched, the difference between experiment groups Lig(83.9±7.7) ,Bai(81.9±6.2) and control group(77.4±5.6) were extremely significant (P<0.01).The development rates of morula cultured for 8-14 hours after frozenthawed in two freezing process, the Bai and the Lig groups were significantly higher than the control group. The livability of Blastocyst cultured for 6-8 hours after frozenthawed in two freezing process, the Bai and the Lig groups were better than the control group.The recipient pregnancy rates, littering rates of embryos, initial weight of baby mouse, mean weight of away cote and living rate of away cote of Bai and Lig groups were better than control group. These results indicated the two Chinese Medicine effective constituents can improve 2-cell mouse embryo quality in vitro culture and make for the developmental potential of frozenthawed transfer embryos.

The projection from nucleus subpre-tectalis/nucleus interstito-pretectalissubpretectalis (SP/IPS) to nucleus rotundus (Rt) is an important part of avian visual pathway. To reveal the morphological features of SP/IPS cells (S cells) which project to Rt, they were retrogradely labeled with cyanine perchlorate DiI in chicks. Two different types of neurons, “type Ⅰ cells” and “type Ⅱ cells” were identified. Based on the amount of the secondary dendrites, these cells were further divided into simple and complex cells, respectively.Morphological analysis indicated that the type I cells were smaller and fewer than type Ⅱcells.
The Anti-abortion Effects of Scutellaria Baicalensis Georgi and Its Components on RU486 Inducing Abortion and CD80+/CD86+ Cells of Spleen in Mice

In order to investigate the significance of the co-stimulatory molecules CD80(B7-1)/CD86(B7-2) in abortion and the anti-abortion mechanisms of Scutellaria baicalensis Georgi and its components, mifepristone (90 μg per mouse) was injected subcutaneously (sc) to induce abortion in BALB/c mice. The number of CD80+ and CD86+ cells in the spleen were assayed by flow cytometry. The results revealed that the CD80+ cells significantly elevated, however, the CD86+ cells significantly decreased in group B (the group treated with RU486 ) in comparison with group A (the control group). The decoction of Scutellaria baicalensis Georgi, and its components (baicalin or baicalein) given orally to the pregnant mice prior to RU486 treatment group significantly inhibited the abortifacient effect of RU486, and its CD80+ cells number significantly decreased, the CD86+ cells number significantly elevated in comparison with the group B. These results suggested that RU486 inducing abortion associated with the CD80+ and CD86+ cells number in the spleen, and that the antiabortion effects of the Scutellaria baicalensis Georgii, its componetnts (baicalin or baicalein) are realized through modulation of the number of CD80+ and CD86+ cells in the spleen.
